Optimasi Konsentrasi Kinetin dan Benzyl Amino Purine Pada Kultur Tunas Vanili (Vanilla planifolia)

Vanili berpotensi untuk dikembangkan melalui teknik kultur jaringan untuk mengantisipasi keterbatasan tanaman induk sebagai sumber bahan tanam. Kemampuan perbanyakan tunas vanili secara in vitro perlu dikendalikan dengan pengaturan Kinetin dan Benzil Amino Purin. Kepentingan penelitian ini adalah 1) analisis respon eksplan vanili pada beberapa konsentrasi Kinetin; 2) analisis respon eksplan vanili pada beberapa konsentrasi BAP dan 3) analisis interaksi Kinetin dan BAP terhadap respon eksplan vanili terhadap perbanyakan tunas. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Politeknik Negeri Jember pada bulan Juni sampai Desember 2020 dengan menggunakan Rancangan Acak Lengkap (RAL) faktorial. Faktor 1 adalah konsentrasi Kinetin 0,0, 1,0, 2,0 mg.L-1 dan faktor kedua adalah konsentrasi BAP 0,5, 1,5, 2,5 mg.L-1. Hasil penelitian membuktikan bahwa perbanyakan tunas paling cepat terjadi pada media MS + Kinetin 2 mg.L-1 dengan rerata 8,7 hari setelah inokulasi. Rerata jumlah pucuk 7,6 pucuk/eksplan dengan berat basah rata-rata tertinggi 0,9 gram/eksplan pada penambahan BAP 1,5 mg. L-1 pada pengukuran 70 hari setelah inokulasi.
